Book excerpt: The progress in protein and nucleic acid chemistry together with improvements of the previously employed tissue culture techniques have led to the solution of problems such as that of the generation of antibody diversity or of the molecular structure of T and B cell membrane receptor for antigen which had challenged the past generations of immunologists. Thanks to this progress an impressive amount of knowledge has been accumulated on certain cell types that were relatively "mysterious" until recently. The B lymphocyte represents a typical example of such a cell. With these considerations in mind, we have started to organize a NATO summer school on "The molecular basis of B cell differentation and function" that had the specific aim of bringing up to date a selected number of young investigators. During the course, held in Santa Margheri ta Ligure, October 1-11, 1985 it became apparent that rather than a formal school we had organized an informal series of Workshops where both the Faculty members and the participants were discussing their own data in addition to reviewing the general progress in the field. The deep commi ttment of everyone to his research subj ect and the enthusiasm had caused this very successful change in the shape of the course. At the end we asked the members of the Faculty and the participants to write a summary of their data and of their point of view on selected subj ect s.

Book excerpt: Rapid progress continues to be made in understanding the molecular and cellular events that comprise B-Iymphocyte differentiation. This is due in part to the high level of inter est shown by many investigators from diverse disciplines, who find this subject suitable for addressing some of the fundamental issues of immunobiology. B-cell developmen tal models are being extensively used to investigate cell-cell interactions, molecular mediators of differentiation and proliferation, differential onset of gene programs, and gene rearrangement and expression, as well as the generation of the immune response itself. Not surprisingly, increased understanding of B-cell differentiation sometimes results from the application of new techniques that permit greater insight into the cells comprising the system and the genetic mechanisms by which these cells express their differentiative potential. However, experimental strategies based upon the novel application of established technologies have also led to the clarification of many issues, as well as to the discov ery of previously unrecognized problems. One problem, well recognized by those active in the field, is how to keep up with significant developments as they appear. The purpose of this book, part of a series devoted to analysing current issues in biology, is to help overcome this problem. No attempt at comprehensive cov erage of all of the issues has been made. Rather, a more thorough analysis of a few topics is presented.

The first chapters review new insights into the assembly of BCR components and their organization on the cell surface. Subsequent contributions focus on the molecular interactions that connect the BCR with major intracellular signaling pathways such as Ca2+ mobilization, membrane phospholipid metabolism, nuclear translocation of NF-kB or the activation of Bruton’s Tyrosine Kinase and MAP kinases. These elements orchestrate cytoplasmic and nuclear responses as well as cytoskeleton dynamics for antigen internalization. Furthermore, a key mechanism of how B cells remember their cognate antigen is discussed in detail. The choice of the best method depends on the biology of the individual malignancy, i.e. on the determination of specific markers which are useful to differentiate between leukemic cells and normal hematopoiesis in leukemic patients. These markers include leukocyte differentiation antigens, fusion transcripts, transcripts overexpressed by mutated or nonmutated genes, rearranged genes, and individual markers like polymorphic repetitive DNA sequences.
