Download or read book Autoradiography and Immunocytochemistry written by M.A. Williams and published by Elsevier. This book was released on 2013-10-22 with total page 238 pages. Available in PDF, EPUB and Kindle. Book excerpt: Autoradiography and Immunocytochemistry sets out in detail the preparation of autoradiographs and methods of preparing and use of immunological staining reagents. This book begins with an introduction to the demand for autoradiography and immunocytochemistry and their most advantageous use, followed by a discussion on the immunocytochemistry at the electron microscopical (EM) level in relation to the preparation of labeled antibodies. Other topics include the general remarks on “staining with antibodies; applying Fer-Ab conjugates to ultrathin sections; and controls in immunoferritin staining. The multistep antibody staining methods with unmodified proteins; control observations in staining experiments using lectins; and pattern analysis in EM immunocytochemistry are also covered. This text concludes with a description of the preparation of electron microscope autoradiographs, including the radioisotopes in EM autoradiography, preparation of radioactively-labeled tissues and cells, and methods of applying nuclear emulsions. This publication serves as a detailed laboratory guide for researchers and workers to successfully conduct electron microscope techniques.

Download or read book Cell Surface Receptors written by Lee E. Limbird and published by Springer Science & Business Media. This book was released on 2013-04-17 with total page 205 pages. Available in PDF, EPUB and Kindle. Book excerpt: The purpose of this book is to serve as a primer for the study of cell surface receptors. The simplified discussion of methods and their underlying prin ciples is intended to remove the usual intimidation caused by the specialized vocabulary or sophisticated mathematics that characterize many of the primary papers in this field. In this way, it is hoped that the basic concepts can be emphasized. This book is meant to be a starting point: a textbook as well as a manual to which the investigator can return for a refresher course, when needed. I feit compelled to write this book for several reasons. The primary philosophical reason was to provide, in one volume, an overall perspective on the study ofcell surface receptors that describes their characterization from pharmacological studies in a whole organ or tissue bath (chapter 2) to studies of radioligand binding in isolated membrane and detergent-solubilized pre parations (chapters 3-5) and, back again, to studies in intact cells or tissues using morphological and biochemical techniques (chapter 6). It is my opinion that the ultimate understanding of the molecular basis for hormone or drug receptor interactions, and the physiological consequences of these inter actions, will require an assessment of cell surface receptor function at all of these experimental levels. In many cases an investigator may not choose to of investigation considered here.

Book Cryotechniques in Biological Electron Microscopy

Download or read book Cryotechniques in Biological Electron Microscopy written by Rudolf A. Steinbrecht and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 308 pages. Available in PDF, EPUB and Kindle. Book excerpt: To preserve tissue by freezing is an ancient concept going back pre sumably to the practice of ice-age hunters. At first glance, it seems as simple as it is attractive: the dynamics of life are frozen in, nothing is added and nothing withdrawn except thermal energy. Thus, the result should be more life-like than after poisoning, tan ning and drying a living cell as we may rudely call the conventional preparation of specimens for electron microscopy. Countless mishaps, however, have taught electron microscopists that cryotechniques too are neither simple nor necessarily more life-like in their outcome. Not too long ago, experts in cryotechniques strictly denied that a cell could truly be vitrified, i.e. that all the solutes and macro molecules could be fixed within non-crystalline, glass-like solid water without the dramatic shifts and segregation effects caused by crystallization. We now know that vitrification is indeed pos sible. Growing insight into the fundamentals of the physics of water and ice, as well as increasing experience of how to cool cells rapidly enough have enlivened the interest in cryofixation and pro duced a wealth of successful applications.
