Book excerpt: There is a prov1s10n in the charter of each Study Section of the Division of Research Grants at the National Institutes of Health that stipulates that "workshops" are to be held periodically to aid Study Section members in their appraisals of recent developments in their fields and to identify future challenges worthy of investigation. The Reproductive Endocrinology Study Section was established on December 13, 1985 to review research grants and research training activities relating to reproductive endocrinology, including aspects of management of reproductive endocrine disorders and hormonal imbalances as related to infertility and during pregnancy and puberty, breast cancer and prostate cancer. It held its first workshop, entitled, "Autocrine and Paracrine Mechanisms in Reproductive Endocrinology," in October, 1988 in Shrewsbury, MA at The Worcester Foundation for Experimental Biology. The proceedings of this workshop, which are detailed herein, reflect the fact that autocrine and paracrine interactions are rapidly being accepted as an exciting area of research by scientists investigating the physiological and biochemical mechanisms of hormone action in the male and female reproductive systems. The material covered is novel and wide-ranging, extending from theoretical considerations of mechanisms of growth factor action and the role of cell cycle stage in determining hormone action to investigations of autocrine and paracrine interactions during development to discussions of the potential clinical ramifications of the basic research findings. Such an extensive inventory is necessary for two reasons.

Book excerpt: Regeneration, the homeostatic ability to maintain tissue structure in the face of normal cell turnover or loss of tissue damaged by trauma or disease, is an essential developmental process that continues throughout life. As recently as a decade ago, any serious discussion of the possibility of regeneration becoming a practical medical tool in the near future had the air of science fiction or over-optimistic speculation. The volume editors believe that an attempt to organize or systematize the literature is long overdue. In this volume, respected experts highlight the latest findings in vertebrate (including mammals) wound healing and regeneration. They present eleven reviews that cover a wide range of topics, from wound repair and its relationship to regeneration, through systems including lenticular, neural, and musculoskeletal tissues and limbs, to epigenetics and the role of the cell cycle. Nuclear reprogramming and cellular plasticity, which open the door for potential regenerative medical therapies for injury and degenerative disease, are recurring themes throughout the book. We are all now part of the regeneration revolution.

Book excerpt: Given that the extremely elaborated and dynamic functions performed by the nervous system require the close synchronization of brain cells, complex organisms have developed different mechanisms of intercellular communication. At this regard, paracrine signaling between neighboring cells is currently recognized as one of the most widely distributed mechanisms of synchronization in the brain parenchyma. In mammals, paracrine signaling is in part mediated by single membrane channels formed by connexins (connexons/hemichannels) or pannexins (pannexons), which are two different membrane protein families composed of about 20 and 3 members, respectively. Single membrane channels formed by these proteins serve as aqueous pores permeable to ions and small molecules, allowing the diffusional exchange between the intra- and extracellular milieu. Thus, connexin hemichannels and pannexons permit the release of significant quantities of autocrine/paracrine signaling molecules (e.g., ATP, glutamate, NAD+, adenosine and PGE2) into the extracellular milieu, as well as the uptake of small molecules. An increasing body of evidence has revealed that connexin hemichannels and pannexons play a crucial role in a plethora of brain processes including blood flow regulation, Ca2+ wave propagation, memory consolidation, glucose sensing and cell migration and adhesion. Considering the multiple cell signaling functions of these channels, their dysregulation is proposed not only as potential pathological biomarker, but it has been implicated in the pathogenesis and progression of diverse brain diseases (e.g., meningitis, Alzheimer’s disease and stroke). The aim of this Research Topic is to gather a collection of original research articles, method, protocols, short communications, opinions, perspectives, as well as review articles, providing the latest progress and insights in the field of connexin hemichannels and pannexons in the nervous system. Within this volume we plan to cover from basic research including channel structure, regulation, pharmacology and trafficking; to different biological functions in the physiology (behavior, plasticity, neurogenesis, blood flow control, neuron-glia crosstalk, cell migration and differentiation) as well as in the pathophysiology (neuroinflammation, mutation-related diseases, glial dysfunction and neurodegeneration) of the nervous system. We hope that this collection of articles will serve to understand how the signaling of connexin hemichannels and pannexons influences both normal and pathological brain function.

As science expands, the emphasis is on data, interpretation, and dogma. We disagree; open minds open new approaches. Using methodologies that are primarily molecular and genetic, we seek to refine the causes of glaucoma as well as how it is best treated, especially incorporating thoughts and hypotheses about new methods of treatment. Glaucoma is a complex disease, and genetics proves that a variety of proteins are culpable at one level. At another level, however, there are likely final common pathways and numerous feedback loops which have defied explanations to date. The search for answers goes on in basic science researcher’s laboratories and clinical ophthalmologist’s offices and operating rooms. We are all well-served by understanding that glaucoma is a neurodegenerative disease. Current attempts to solve the disease have focused on two strategic arenas: the trabecular meshwork function and its impact on intraocular pressure as a major risk factor for the disease; and the optic nerve dysfunction leading to visual loss. Genetic mutations have yielded puzzling clues to the cause, but without resolution. For example, mutations in myocilin and optineurin genes are closely connected to the phenotype, but how do they cause the disease? In the next two years, priority areas of research are signaling pathway discoveries, biomarker panels, epigenetic factors, and continued genomic studies to yield answers to the common final pathways of the disease. The final pathways are complex and redundant, such that the overlap of bio-informatics will be challenging. Current promising leads suggest the innate immune system holds important clues to both trabecular meshwork and optic nerve pathophysiology. When the primary open-angle glaucoma disease pathways are unraveled, drug discoveries and new treatment modalities will be available for better regulation of intraocular pressure and neuroprotection for the optic nerve. This volume discusses the glaucoma pipeline from several perspectives as well as future candidate classes. Book excerpt: The skin, the body’s largest organ, is strategically located at the interface with the external environment where it detects, integrates and responds to a diverse range of stressors, including solar radiation. It has already been established that the skin is an important peripheral neuroendocrine-immune organ that is closely networked with central regulatory systems. These capabilities contribute to the maintenance of peripheral homeostasis. Specifically, epidermal and dermal cells produce and respond to classical stress neurotransmitters, neuropeptides and hormones, production which is stimulated by ultraviolet radiation (UVR), biological factors (infectious and non-infectious) and other physical and chemical agents. Examples of local biologically active products are cytokines, biogenic amines (catecholamines, histamine, serotonin and N-acetyl-serotonin), melatonin, acetylocholine, neuropeptides including pituitary (proopiomelanocortin-derived ACTH, b-endorphin or MSH peptides, thyroid stimulating hormone) and hypothalamic (corticotropin-releasing factor and related urocortins, thyroid-releasing hormone) hormones, as well as enkephalins and dynorphins, thyroid hormones, steroids (glucocorticoids, mineralocorticoids, sex hormones, 7-δ steroids), secosteroids, opioids and endocannabinoids. The production of these molecules is hierarchical, organized along the algorithms of classical neuroendocrine axes such as the hypothalamic pituitary adrenal axis (HPA), hypothalamic-thyroid axis (HPT), serotoninergic, melatoninergic, catecholaminergic, cholinergic, steroid/secosteroidogenic, opioid and endocannabinoid systems. Disruptions of these axes or of communication between them may lead to skin and/or systemic diseases. These local neuroendocrine networks also serve to limit the effect of noxious environmental agents to preserve local and consequently global homeostasis. Moreover, the skin-derived factors/systems can also activate cutaneous nerve endings to alert the brain to changes in the epidermal or dermal environments, or alternatively to activate other coordinating centers by direct (spinal cord) neurotransmission without brain involvement. Furthermore, rapid and reciprocal communications between epidermal and dermal and adnexal compartments are also mediated by neurotransmission including antidromic modes of conduction. Lastly, skin cells and the skin as an organ coordinate and/or regulate not only peripheral but also global homeostasis.

Book excerpt: C1q is the target recognition protein of the classical complement pathway and a major connecting link between innate and acquired immunity. As a charge pattern recognition molecule of innate immunity, C1q can engage a broad range of ligands derived from self, non-self and altered self via its heterotrimeric globular (gC1q) domain and thus trigger the classical complement pathway. The trimeric gC1q signature domain has been identified in a variety of non-complement proteins that can be grouped together as a C1q family. C1q circulates in serum as part of the C1 complex, in association with a catalytic tetrameric assembly of two homologous yet distinct serine proteases, C1r and C1s. Binding of C1q to appropriate targets leads to sequential activation of C1r and C1s, the latter being able to cleave complement components C4 and C2 thereby triggering the complement cascade. Activation of the classical pathway plays an important role in innate immune protection against pathogens and damaged elements from self. However, its involvement has been shown in various pathologies including ischemia-reperfusion injury and hereditary angioedema. Unexpected roles for the classical pathway have also been discovered recently, linked to both physiological and pathological aspects of development, including brain and cancer cells. These new perspectives should arouse renewed interest in a search for specific inhibitors of the classical pathway. In addition, C1q has recently been shown to have a number of functions that are independent of the activation of the classical pathway. This research topic is aimed at providing a state-of-the-art overview of the classical pathway, including, but not restricted to emerging functions of C1q and of the C1 complex, as well as pathological consequences of C1 activation or of the presence of anti-C1q autoantibodies . Contributions are included in the areas such as structural basis of C1q ligand recognition, C1q family proteins, inhibitors of the classical pathway identified in pathogens and improved derived inhibitors, structural determinants of the substrate specificities of C1r and C1s, elucidation of the architecture of C1, structural and functional homology of C1 with the initiating complexes of the lectin complement pathway, and novel involvement of C1q in processes such as ageing, cancer, synaptic pruning, and pregnancy.
