Book excerpt: Chiral lithium amide bases (CLAB's) have been successfully used in asymmetric organic reactions in the past decades. Despite the increasing use of polymeric reagents in organic synthesis, the number of papers describing polymer-supported chiral lithium amide bases (pCLAB's) is limited. The pCLAB's have several advantages such as easy separation and reuse. Additionally, the "polymeric effect" often plays an important role in organic reactions, such as physical separation of aggregates and steric interference. All these prompted us to study the structure and reactions of pCLAB's. Polymer-supported chiral amines were effectively prepared from amino acid derivatives and Merrifield resin. Treatment of polymer-supported amines with n-butyllithium gave the corresponding pCLAB's which were tested in asymmetric deprotonation reactions of prochiral ketones. Trimethylsilyl enol ethers were obtained in up to 82% ee at room temperature without additives. The polymer-supported chiral lithium amides can be readily recycled and reused without significant loss of reactivity and selectivity.

Book excerpt: 6Li and 15N NMR spectroscopic investigations of lithium diisopropylamide-mediated ester enolization in THF, t-BuOMe, HMPA/THF, and DMPU/THF reveal substantial concentrations of mixed aggregates. While quantitative rate studies revealed that the metalations by lithium diisopropylamide homonuclear dimers proceed at nearly solvent-independent rates, the reactivities of the intermediate mixed aggregates are markedly lower and quite sensitive to the choice of solvent. The autoinhibition correlates the relative stabilities of the mixed aggregates. However, the relative stabilities do not correlate in a simple fashion with the ligating properties of the solvent.
