Book Solution and Solid Phase Synthesis of Isoxazoline Containing Heterocycles

Download or read book Solution and Solid Phase Synthesis of Isoxazoline Containing Heterocycles written by Kristin Ann Milinkevich and published by . This book was released on 2009 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: An efficient, multi-gram synthesis of a spiroisoxazolinoproline-based amino acid requiring minimal purification, delivering good cis:trans diastereoselectivity (ca. 1:4) and providing good yields is reported. Surface bound studies of the reduction of an aryl-nitro group in the presence of an isoxazoline ring with tin(II) dichloride dihydrate were undertaken to confirm the stability of the isoxazoline ring. Full derivitization of this spiroisoxazolinoproline-based amino acid scaffold was performed during the synthesis of a library. This work is described in Chapter 1. Condensation of 3-chloropentane-2,4-dione with thioamides gives 1-(thiazol-5-yl)ethanones and subsequent Wittig olefination followed by nitrile oxide 1,3-dipolar cycloaddition to the resulting prop-1-en-2-yl moiety delivers racemic 5-(thiazol-5-yl)-4,5-dihydroisoxazoles. Biological evaluation of these diheterocycles showed promising fungicidal activity. This work is described in Chapter 2. A route to 2-(4,5-dihydroisoxazol-5-yl)-1,3,4-oxadiazoles has been developed. Acid hydrazide starting materials underwent an EDC-mediated coupling with acrylic acid derivatives to form diacyl hydrazides. Cyclodehydration of the diacyl hydrazide in the presence of triphenylphospine and hexachloroethane gave 1,3,4-oxadiazoles. Lastly, in situ nitrile oxide formation and subsequent 1,3-dipolar cycloaddition to the exomethylene moiety delivered 2-(4,5-dihydroisoxazol-5-yl)-1,3,4-oxadiazoles. Biological evaluation of these diheterocycles showed weak insecticidal activity. This work is described in Chapter 3. A library of novel, propeller-shaped dispirotriheterocyclic isoxazolinopiperidinochromanones is reported. Each rigid dispirotriheterocycle was prepared in five linear steps from commercially available tert-butyl 4-oxopiperidine-1-carboxylate plus various derivatives of 1-(2-hydroxyphenyl)ethanone, benzaldehyde oxime, and carboxylic acids. Computational chemistry was employed to analyze the three-dimensional geometries of these dispirotriheterocycles as well as to generate chemoinformatic bioavailability data. X-ray crystallographic structure determination verified the regioselectivity of the nitrile oxide 1,3-dipolar cycloaddition reaction. This work is described in Chapter 4. A route to spiroisoxazolinopiperidinylbenzamides has been developed. N-Boc-4-piperidone underwent a Wittig olefination and Boc deprotection followed by an ipso substitution with 4-fluoro-3-nitrobenzoic acid to yield the starting scaffold in excellent yields. Diversification of the acid with primary amines, followed by in situ nitrile oxide formation and subsequent 1,3-dipolar cycloaddition to the exomethylene moiety delivered the spiroisoxazolinopiperdine intermediates. Reduction of the arylnitro followed by acylation or reductive amination yielded the spiroisoxazolinopiperidinylbenzamide library. This work is described in Chapter 5.

Available in PDF, EPUB and Kindle. Book excerpt: Modern Applications of Cycloaddition Chemistry examines this area of organic chemistry, with special attention paid to cycloadditions in synthetic and mechanistic applications in modern organic chemistry. While many books dedicated to cycloaddition reactions deal with the synthesis of heterocycles, general applications, specific applications in natural product synthesis, and the use of a class of organic compounds, this work sheds new light on pericyclic reactions by demonstrating how these valuable tools elegantly solve synthetic and mechanistic problems. The work examines how pericyclic reactions have been extensively applied to different chemistry areas, such as chemical biology, biological processes, catalyzed cycloaddition reactions, and more. This work will be useful for organic chemists who deal with organic chemistry, medicinal chemistry, agrochemistry and material chemistry. - Provides details on the synthesis of antiviral and anticancer compounds, marking the key role of unconventional catalyzed cycloaddition reactions for preparing new derivatives in a unique reaction pathway that is scalable in industrial processes - Contains the most up-to-date review of the use of pericyclic reactions in drug delivery - Includes the enzyme-catalyzed processes involving cycloaddition reactions for different targets, demonstrating that cycloaddition is more common in nature than expected - Features new applications for cycloadditions in material chemistry and provides a general view of the most recent results in the area

Book Organic Synthesis with Carbohydrates

Download or read book Organic Synthesis with Carbohydrates written by Geert-Jan Boons and published by John Wiley & Sons. This book was released on 2008-04-15 with total page 348 pages. Available in PDF, EPUB and Kindle. Book excerpt: Carbohydrates offer a ready source of enantiomerically pure starting materials. They have been used for the imaginative synthesis of a wide range of compounds, and have been found to be effective chiral auxiliaries which enable the introduction of a range of functionalities in a highly enantioselective manner. In a subject dominated by volumes at research and professional level, this book provides a broad understanding of the use of carbohydrates in organic synthesis, at postgraduate student level. Emphasis is placed on retrosynthetic analysis, with discussion of why a particular synthetic route has been chosen, and mechanistic explanations are provided for key and novel reactions. Wherever possible, the authors highlight points of general significance to organic synthesis. Selected experimental conditions and reaction details are incorporated to ensure that information can be utilised in research. The book is extensively referenced and so provides a convenient point of entry to the primary literature.

Book excerpt: Carbon analogs of carbohydrates, dubbed C-glycosides, have remained an important and interesting class of mimetics, be it in natural product synthesis, for pharmacological applications, as conformational probes, or for biological studies. C-Furanosides: Synthesis and Stereochemistry provides a much-needed overview of synthetic and stereochemical principles for C-furanosides: analogs of a 5-membered ring carbohydrate glycoside (furanoside), in which the anomeric oxygen has been replaced with a carbon. While our understanding of conformational behavior and of stereoselective synthesis in 6-membered ring compounds is quite good, our ability to predict the conformation of 5-membered ring compounds, or to predict the stereochemical outcome of a given reaction, remains anecdotal. Through a comprehensive review of literature approaches to the different C-furanoside stereoisomers, as well as an interpretation of the outcome in terms of a reasonable number of stereochemical models, C-Furanosides: Synthesis and Stereochemistry enables the reader to determine the best approach to a particular C-glycoside compound, and also hopes to provide a certain level of rationalization and predictability for the synthesis of new systems. - Provides a comprehensive review of the growing literature in C-furanosides - Enables readers to choose the most convenient approach to access a defined target in natural products synthesis or pharmacology and make reasonable predictions for the stereochemical outcome in unpublished cases - Explores the various rational models for stereochemical analysis of furanoside reactivity, with a clear distinction made between physical chemical mechanisms and stereochemical models

Download or read book Carbohydrate Mimics written by Yves Chapleur and published by Wiley-VCH. This book was released on 1998-09-10 with total page 638 pages. Available in PDF, EPUB and Kindle. Book excerpt: Carbohydrate Mimics Concepts and Methods Edited by Yves Chapleur Apart from the various functions of carbohydrates in natural processes they are important building blocks for drugs, especially antibiotics. In addition, carbohydrates are considered as important structures in the immunological recognition process of the antigen-antibody reaction. The modification of the structure and thereby the chemistry of substrates is a common procedure in nature. This naturally occurring process is reflected in the concept of molecular mimicry, which has become a useful approach in drug discovery. The search for carbohydrate analogues that exhibit better binding activity, biological activity, and stability began with the synthesis of azasugars. A lot of structural modifications around the anomeric center of carbohydrates has been achieved over the last decade, and these are summarized in terms of the different chemical and biological aspects of carbohydrate mimics in one part of this book. In addition to the well established fields of C-glycosides, azasugars and carbasugars the other part of the book focusses on outstanding achievements in carbohydrate mimic research. Everybody who is active in carbohydrate research or is interested in moving into this exciting field will benefit from this up-to-date source of informations. The leading experts in this field describe their scientific approaches to real interdisciplinary puzzles. This book should be an inspiration to scientists in organic, medicinal chemistry, biochemistry, and biology.
