Book excerpt: Since the appearance of the first two volumes of Modern Fluorescence Spectroscopy in 1976, important advances continue to be made in both the techniques and applications of molecular luminescence. In terms of "hardware," it is only recently that the application of laser excitation to molecular fluorometry has become feasible under conditions that are analy tically realistic. The improvements that can be effected in sensitivity, analy tical selectivity, and ability to handle "difficult" samples by laser fluorometry have only begun to be exploited. Likewise, time-resolved fluorometry has received widespread use in fundamental studies (a sizable number of which deal with biological systems), but has as of yet received relatively little analytical utilization. The use of electronic array detectors offers the promise of obtaining luminescence spectra more rapidly, and perhaps ultimately with greater sensitivity, than is possible by the use of scanning instruments equipped with conventional detectors. The increasing capabilities of microcomputers and the increasing sophistication of "smart" spectroscopic instrumentation signify that much more efficient acquisition and use can now be achieved of the information contained in the "excitation-emission matrix" inherent in the luminescence phenomenon.

Book excerpt: Laser induced fluoresence (LIF) spectra (laser excitation spectra) are conceptually among the most simple spectra to obtain. One need only confine a gaseous sample in a suitable container, direct a laser along one axis of the container, and monitor the sample's fluorescence at a right angle to the laser beam. As the laser wavelength is changed, the changes in fluorescence intensity map the absorption spectrum of the sample. (More precisely, only absorption to states which have a significant radiative decay component are monitored.) For ion spectroscopy, one could benefit in many ways by such an experiment. Most optical ion spectra have been observed by emission techniques, and, aside from the problems of spectral analysis, discharge emission methods often produce the spectra of many species, some of which may be unknown or uncertain. Implicit in the description of LIF given above is certainty as to the chemical identity of the carrier of the spectrum. This article describes a method by which the simplifying aspects of LIF can be extended to molecular ions (albeit with a considerable increase in experimental complexity over that necessary for stable neutral molecules).

Book excerpt: Low-temperature laser-induced fluorescence spectroscopy has various applications in analytical, physical, and biophysical chemistry. This technique provides information on the fluorescence origin band, zero-phonon lines and phonon-sidebands, inhomogeneous broadening, electron-phonon coupling strength, and ground- and excited-state vibrational frequencies of studied molecules. Examples discussed in this work include studies of DNA/metabolites and monoclonal antibody (mAb)/antigen interactions. The structural basis for the increased reactivity of BPDE towards guanines at 5-methylcytosine ([superscript]M[superscripte]eC):G sites in DNA was investigated by low temperature laser-based spectroscopy, studying the nature of physical complexes of benzo[a]pyrene tetraol in a series of 5-methylcytosine structural DNA analogs. We found that the presence of a C-5 substituent on cytosine and related structural modifications influences the conformation of BPT in DNA analogs, and could explain the increase in guanine reactivity at [superscript]M[superscript]eC:G sites of the p53 tumor suppressor gene that contains endogenenous 5-([superscript]M[superscript]eC. It has been demonstrated that various mAbs can bind a particular cross-reactant by adopting two distinct "red" and "blue" conformations of its binding sites. We showed that the blue conformation of pyrene in several mAbs (including 4D5 mAb) is consistent with [pi]-cation interactions, underscoring the importance of [pi]-cation interaction in ligand binding. We propose that considerable narrowing of the fluorescence origin band of the ligand in the protein environment could be regarded as a simple indicator of [pi]-cation interactions. It is also shown that time-resolved delta fluorescence line-narrowing ([delta]FLN) spectroscopy, using excitation within the (0,0)-transition band, provides more reliable information of the frequency dependence of the electron-phonon coupling (Huang-Rhys factor, (S
