Book excerpt: The studies of free radicals in melanin and the human melanoma malignum cells by an X-band (9.3 GHz) electron paramagnetic resonance (EPR) spectroscopy were presented. The original results were compared with those published earlier. The aim of this work was application of the advanced spectral analysis to determine free radical properties in melanin biopolymers obtained from different melanotic tumor cells and free radicals existing in the human melanoma cells. Magnetic spin-lattice interactions in melanin samples were tested. The evolution of lineshape of tumor cells with increasing of microwave power was determined to confirm their complex free radical system. The useful shape parameters were proposed. The shape of melanotic tumor cells was analyzed. EPR spectra of free radicals in the melanin isolated from different tumor cells measured in the wide range of microwave power were analyzed. The melanins were obtained from the control tumor cells and the cells cultured with the several antitumor substances. The usefulness of the electron paramagnetic resonance spectroscopy was confirmed.

Abstract: Abstract of thesis entitled Analysis of Free Radical Characteristics in Biological Systems Based on EPR Spectroscopy, Employing Blind Source Separation Techniques Submitted by Ren Jiyun for the degree of Doctor of Philosophy at The University of Hong Kong in June 2006 Free radicals, such as nitric oxide (NO), are highly reactive molecules that have been implicated critical in many physiological and pathophysiological pathways. Electron paramagnetic resonance (EPR) is recognized as a direct and reliable technique to measure a free radical, but the precise measurement of a free radial is still difficult to achieve. This thesis is therefore focused on the following two subjects. The first objective of the study is to find a proper method that can accurately separate the 'pure' components from mixed EPR spectra, as EPR spectrum of a free radical is often superimposed by overlapping spectra of other species. Two blind source separation (BSS) techniques, one based on independent component analysis (ICA), and the other based on sparse component analysis (SCA), were for the first time applied to EPR spectroscopy analysis. The studies, on both computer simulated free radical spectra and biologically obtained signals, show that both BSS techniques could give good separation provided that the source component spectra lines are not very broad and there are prominent non-overlapping peaks. The implementation of this method is simple, automatic, efficient, and does not require the component spectra to be known in advance. The novel method demonstrated a big improvement to the traditional means of manual work or with more sophisticated biomedical experiments. Moreover, compared to the automatic PCA and self-modeling techniques which have previously been used to separate complex EPR spectra, this study confirms that the two BSS techniques are more accurate, robust to noise, and simpler to implement in the cases of three and even more components. Within the two BSS approaches, the SCA approach, which is novelly developed exploiting the sparse property of signals, achieves even more accurate results. With the help of BSS techniques to extract the pure NO signal from the measured mixture signal, the second objective in the present study is to investigate experimentally the NO radical level which helps interpret the possible actions and mechanisms of NO during renal ischemia-reperfusion (I/R) of rats. When EPR is used to detect NO production, the two NO spin-trapping methods, "internal" hemoglobin trapping and "external" DETC-Fe trapping, result in two NO spin adducts, namely 2+ DETC -Fe -NO and HbNO. This thesis presents a novel comparison study on the properties of the two spin-trapping methods. Renal I/R model on rats were established through occluding the left renal artery for 30 min or 60 min. Animal experiments show that the external DETC-Fe trapping method is more sensitive, but 2+ DETC -Fe -NO in kidney would quickly decompose, thus can only be used for short-term I/R injury study; whereas the internal hemoglobin trapping is not so sensitive to small amount of NO, but more stable and tend to accumulate in blood, hence suitable for long-term in vivo I/R injury study. The majority of natural dyes are vegetable dyes from plant sources. Dyeing is the process of imparting colors to a textile material. Different classes of dyes are used for different types of fiber and at different stages of the textile production process, from loose fibers through yarn and cloth to completed garments. There are technologies that manufacture the pigments for plastics, rubber and cosmetics. Therefore; dyes and pigments have a vast area of applications and have a huge demand in industry. Contrary to popular opinion, natural dyes are often neither safer nor more ecologically sound than synthetic dyes. They are less permanent, more difficult to apply, wash out more easily, and often involve the use of highly toxic mordant. Of course, the colour possibilities are far more limited; the color of any natural dye may be easily copied by mixing synthetic dyes, but many other colors are not easily obtained with natural dyes. However, some mordant are not very toxic, and the idea of natural dyestuffs is aesthetically pleasing. Applying natural dyes in your fabric production using enzymes will reduce your production cost and improve control. There are various kind of natural dyes; quinonoid dyes, cyanine dyes, azo dyes, biflvonyl dyes, omochromes, anthraquinone, coprosma gesus etc. The use of natural dyes in cloth making can be seen as a necessary luxury to trigger off a change in habits. Dyes which stand out for their beauty and ecological attributes would never be employed on just any material but on noble fabrics such as wool, silk, linen or cotton, made to last more than one season. Market value will benefit from consumer preferences for environmentally friendly products, which will support consumption of high performance dyes and organic pigments. This book describes various sample-related, instrument-related and software-related aspects of obtaining quantitative results from EPR expe- ments. Some speci?c items to be discussed include: selection of a reference standard, resonator considerations (Q, B ,B ), power saturation, sample position- 1 m ing, and ?nally, the blending of all the factors together to provide a calculation model for obtaining an accurate spin concentration of a sample. This book might, at ?rst glance, appear to be a step back from some of the more advanced pulsed methods discussed in recent EPR texts, but actually quantitative “routine CW EPR” is a challenging technique, and requires a thorough understa- ing of the spectrometer and the spin system. Quantitation of CW EPR can be subdivided into two main categories: (1) intensity and (2) magnetic ?eld/mic- wave frequency measurement. Intensity is important for spin counting. Both re- tive intensity quantitation of EPR samples and their absolute spin concentration of samples are often of interest. This information is important for kinetics, mechanism elucidation, and commercial applications where EPR serves as a detection system for free radicals produced in an industrial process. It is also important for the study of magnetic properties. Magnetic ?eld/microwave frequency is important for g and nuclear hyper?ne coupling measurements that re?ect the electronic structure of the radicals or metal ions.
