Book excerpt: "Biologically Based Methods for Cancer Risk Assessment", an Advanced Research Workshop, (ARW) sponsored by the North Atlantic Treaty Organization (NATO) was held in Corfu, Greece in June, 1989. The intent of the workshop was to survey available pharmacokinetic and pharmacodynamic methods in cancer risk assessment and identify methodological gaps and research needs for biologically based methods in cancer risk assessment. Incorporation of such methods represents one of the most challenging areas for risk assessment. The workshop included an international group of invited experts in the field and provided for a dynamic exchange of ideas and accomplishments. Some of the major topics discussed were: * Inventory of available pharmacokinetic and pharmacodynamic methods for cancer risk assessment. * Identification of methodology gaps and research needs in biologically based methods in cancer risk assessment. * Development of a general framework to guide future cancer risk assessment research. This book is a compilation of the papers presented at the workshop and is intended to provide guidance for future research to reduce uncertainties in the cancer risk assessment process. The primary sponsorship of this ARW by NATO and the advice and cooperation of Dr. C. Sinclair of the Scientific affairs Division are gratefully acknowledged. These Proceedings are a result of the eleventh conference of Scientific Issues in Quantitative Cancer Risk Assessment, which was held in 1989 at Snowbird, Utah. For five days, 45 speakers and observers contributed their expertise in the relevant biology and statistics. The presentations were timely and the discussion was both enlightening and at times spirited. This volume hopefully presents a record that will be useful both now and in the future. Suresh H. Book excerpt: An integrated retrovirus effectively becomes part of the cellular genome, but with the difference that the virus to a large extent retains control over its own expression through nontranslated sequences in the long terminal repeat (L TR). Some retroviruses also code for nonstructural proteins that further regulate proviral expression. Integration changes the cell genome; it adds viral genes, and in the case of transducing retroviruses also adds cell-derived oncogenes that have been incorporated into the viral genome. Integration can also have consequences for cellular genes. The transcriptional signals in a provirus can activate expression of neighboring cellular genes; the integration even can disrupt and thus inactivate cellular genes. These effects of retroviral genomes take place in cis; they are referred to as insertional mutagenesis and are the subject of this volume. Almost 10 years have passed since W. Hayward, S. Astrin, and their colleagues found that in B cell lymphomas of chickens, induced by avian leukosis virus, transcription of the cellular proto-oncogene myc was upregulated through the integration of a complete or partial provirus in its vicinity. This landmark discovery suggested a mechanism by which retro viruses that do not carry cellular oncogenes in their genome ("nonacute retroviruses") can cause cancer. It contributed the first evidence for the carcinogen potential of oncogenes that are not part of a viral genome.
