Certaines toxines végétales depuis longtemps utilisées figurent encore dans la pharmacopée actuelle. Viennent s'y ajouter depuis peu des toxines microbiennes. Cependant le maniement des toxines natives ou à peine modifiées reste délicat tant est faible la marge entre les effets toxiques et l'efficacité thérapeutique. Comment, dans une perspective médicale, canaliser la puissance d'action et l'efficacité d'une toxine ? Connaître sa structure, comprendre son mécanisme d'action n'est pas suffisant : il faut aussi identifier sa cible privilégiée, autrement dit un récepteur cellulaire, faute de quoi l'entreprise apparemment chimérique d'un détournement de sa puissance et de sa spécificité reste aveugle. Ce cap est franchi, depuis un certain nombre d'années, et le développement des recherches sur les toxines axées sur la thérapeutique est devenu foisonnant. Voyez telle ou telle toxine protéique ternaire, avec ses trois domaines, de fixation sur un récepteur membranaire, de translocation qui lui permet de franchir la membrane cellulaire, et d'activité enzymatique qui va alors pouvoir s'exercer dans la cellule. Qu'on parvienne à dissocier chacun de ces trois domaines en leur conservant leurs propriétés, et des possibilités nouvelles d'utilisation thérapeutique vont s'ouvrir, presque à l'infini semble-t-il. Car chacun d'entre eux pourra être couplé à une molécule active bien ciblée, et leur efficacité sera même améliorée pour peu qu'on parvienne à conjuguer leur action. Qu'un récepteur, qu'une molécule, propres au métabolisme ou à la physiologie d'une cellule cancéreuse soient identifiés, et des toxines conjuguées à un ligand de ce récepteur, de cette molécule (ce peut être un anticorps) bloqueront les canaux ioniques ou exerceront leur effet cytolytique sur cette seule cellule, sans nuire aux cellules saines. Nous voilà dégagés de l'utopie, le contenu de cet ouvrage le prouve abondamment. Et même si la concrétisation des promesses reste délicate, le champ d'action est si vaste initialement qu'il ne saurait rester improductif : il ne l'est d'ailleurs déjà plus.

The scientific program included plenary lectures and symposia, in addition to poster presentations and colloquia. The abstracts of the Congress presentations were published as .supplement 1 of volume 72 by the Cana dian Journal of Physiology & Pharmacology. The Congress organizers sought a more expansive treatment of the Congress proceedings and appointed Dr. A. Claudio Cuello to coordinate preparation of the present volume; Dr. Brian Collier was chair of the scientific program committee and, thus, also collaborated on this work. The objective that we pursued was to produce a volume of reasonable size which would feature all of the plenary lectures and symposia from those authors who agreed to participate. To this end, we solicited mini reviews from plenary lecturers and asked symposia organizers to coordi nate a single short-review covering the individual topics within their event. Those who accepted this challenge are evident in this volume. We express our gratitude to these authors for doing so, and for exercising considerable ingenuity in completing their task within a reasonable time.

Inversement, les maladies systémiques sont l’objet de nombreuses interrogations de la part des ophtalmologistes. De ce constat est né Œil et Maladies systémiques qui présente, en 50 chapitres : • les principaux syndromes ophtalmologiques et les moyens d’explorations, destinés principalement aux non-ophtalmologistes • l’expression ophtalmologique des maladies systémiques (connectivites et vascularites, certaines maladies infectieuses et autres affections multiviscérales) ainsi que les aspects diagnostiques, pronostiques et thérapeutiques. Ovarian cancer is cancer that begins in the cells that constitute the ovaries, including surface epithelial cells, germ cells, and the sex cord-stromal cells. Cancer cells that metastasize from other organ sites to the ovary (most commonly breast or colon cancers) are not then considered ovarian cancer. According to the American Cancer Society, ovarian cancer accounts for 4 percent of all cancers among women and ranks fifth as a cause of their deaths from cancer. The American Cancer Society statistics for ovarian cancer estimate that there will be 25,400 new cases and 14,300 deaths in 2003. The death rate for this disease has not changed much in the last 50 years. Unfortunately, almost 70 percent of women with the common epithelial ovarian cancer are not diagnosed until the disease is advanced in stage -- i.e., has spread to the upper abdomen (stage III) or beyond (stage IV). The 5-year survival rate for these women is only 15 to 20 percent, whereas the 5-year survival rate for stage I disease patients approaches 90 percent and for stage II disease patients approaches 70 percent. Ovarian tumors are named according to the type of cells the tumor started from and whether the tumor is benign or cancerous. The three main types of ovarian tumors are: Epithelial Tumors, Germ Cell Tumors and Stromal Tumors.
