Currently, several different cancer vaccine formulations such as peptides, proteins, antigen-pulsed dendritic cells, whole tumor cells, etc. in combination with various adjuvants and carriers are being evaluated in clinical trials (1-3). To determine the optimal cancer vaccine strategy, a surrogate immunological end-point that correlates with clinical outcome needs to be defined, since it would facilitate the rapid comparison of these various formulations. Traditional immunological assays such as ELISA, proliferation and cytotoxicity assays can detect immune responses in vaccinated patients but are not quantitative. In contrast, novel assays such as enzyme-linked immunospot (ELISPOT) assay, intracellular cytokine assay and tetramer assay can quantitate the frequency of antigen-specific T cells. Of these, the ELISPOT assay has the 5 lowest detection limit with 1/10 peripheral blood mononuclear cells (PBMC) and has been determined to be one of the most useful assays to evaluate immune response to cancer vaccines (4). However, the IFN-? ELISPOT assay is not an exclusive measure of cytotoxic T-lymphocyte (CTL) activity as non-cytotoxic cells can also secrete IFN-?. Additionally, CTL with lytic activity do not always secrete IFN-? (5). A more relevant approach to assess functional activity of cytotoxic lymphocytes would be to measure the secretion of molecules that are associated with lytic activity. One of the major mechanisms of cell-mediated cytotoxicity involves exocytosis of cytoplasmic granules from the effector toward the target cell.

Book excerpt: “Infogest” (Improving Health Properties of Food by Sharing our Knowledge on the Digestive Process) is an EU COST action/network in the domain of Food and Agriculture that will last for 4 years from April 4, 2011. Infogest aims at building an open international network of institutes undertaking multidisciplinary basic research on food digestion gathering scientists from different origins (food scientists, gut physiologists, nutritionists...). The network gathers 70 partners from academia, corresponding to a total of 29 countries. The three main scientific goals are: Identify the beneficial food components released in the gut during digestion; Support the effect of beneficial food components on human health; Promote harmonization of currently used digestion models Infogest meetings highlighted the need for a publication that would provide researchers with an insight into the advantages and disadvantages associated with the use of respective in vitro and ex vivo assays to evaluate the effects of foods and food bioactives on health. Such assays are particularly important in situations where a large number of foods/bioactives need to be screened rapidly and in a cost effective manner in order to ultimately identify lead foods/bioactives that can be the subject of in vivo assays. The book is an asset to researchers wishing to study the health benefits of their foods and food bioactives of interest and highlights which in vitro/ex vivo assays are of greatest relevance to their goals, what sort of outputs/data can be generated and, as noted above, highlight the strengths and weaknesses of the various assays. It is also an important resource for undergraduate students in the ‘food and health’ arena.

In the last century, it has become clear that in patients suffering certain types of cancer, spontaneous activation of the immune sytem may lead to curation. The concept of immunotherapy is based on that notion. The intention is to stimulate T cells directed against the cancer, leading to the eradication of cancer cells. However, the methods to stimulate T cell immunity are just evolving, and at the moment they are functional in small laboratory animals only. Furthermore, we know now that many cancer cells possess the capacity to scive attacks of the immune system. This thesis introduces a novel T cell vaccination method that uses a tattoo machine to inject DNA in the skin of the vaccinee. In comparison to other experimental vaccination methods DNA tattooing is very strong: besides small laboratry animals also large animals (mockeys) mount strong T cell responses on tattoo DNA vaccination. Future tests in melanoma patients will point out whether DNA tattoo vaccination is equally efficient in humans, and whether it may have a therapeutic effect in these patients. In this context, T cells play the major role in cell mediated adaptive immune response. The protective role of CD4+ and CD8+ T cells has been inferred from studies on patients who recovered from SARS and MERS and accumulating data are now showing their relevance in SARS-CoV-2 infection. Moreover, memory T cells induced by previous pathogens can shape the susceptibility to, and the clinical severity of other infections, but the complete picture has yet to be elucidated. If the virus is not rapidly eliminated, COVID-19 may progress towards a secondary inflammatory phase that is directly responsible for a worsening in clinical symptoms and immune system impairment. Besides marked lymphopenia, COVID-19 patients’ T cell compartment displays several alterations involving different subpopulations of T cells in terms of phenotype, metabolic profile and functionality.
