Book excerpt: Human exposure to polycyclic aromatic hydrocarbons (PAHs) due to cigarette smoke is well known. It is also known that PAHs are not inherent components of tobacco. Their presence in cigarette smoke results from the curing process of tobacco leaves used for the fabrication of cigarettes and the incomplete combustion of tobacco during the smoking process. The two chromatographic versions of EPA method 610 are popular approaches for the analysis of PAHs in a wide variety of samples. This method, which is based either on High-Performance Liquid-Chromatography (HPLC) or Gas-Chromatography (GC) has shown the presence of Benzo[a]pyrene (B[a]P) in tobacco samples and cigarette smoke. B[a]P is the most toxic PAH in the U.S Environmental Protection Agency (EPA) priority pollutant list.

Book excerpt: Studies of phase distributions and emission factors for polycyclic aromatic hydrocarbons (PAH) in environmental tobacco smoke (ETS) require collection and analysis of very small samples. To achieve the necessary selectivity and sensitivity, a method has been devised and tested for extraction and cleanup of gas- and particulate-phase ETS samples. Gas-phase species were trapped by polymeric sorbents, and particles were trapped on filters. The samples were extracted with hot cyclohexane, concentrated and passed through silica solid-phase extraction columns for cleanup. After solvent change, the PAH were determined by high performance liquid chromatography with two programmed fluorescence detectors. PAH concentrations in 15-mg aliquots of National Institute of Standards and Technology Standard Reference Material SRM 1649 (Urban DustIOrganics) agreed well with published values. Relative precision at the 95% confidence level was 8% for SRM 1649 and 20% for replicate samples (5 mg) of ETS particles. Emission factors have been measured for a range of gas- and particulate-phase polycyclic aromatic hydrocarbons in ETS. The emission factors per cigarette were 13.0±0.5 mg particulate matter, 11.2±0.9 pg for gas-phase naphthalene and 74±10 [mu]g for particulate benzo(a)pyrene.

A classic example is the analysis of the sixteen polycyclic aromatic hydrocarbons included in the priority pollutants list of the U.S. Environmental Protection Agency (EPA-PAHs).

Book excerpt: An analytical method is described for the simultaneous quantitation of polycyclic aromatic hydrocarbons [pAHs] and alkylderivatives [alkyl-PAHs] in 'real world' samples using microbore reversed-phase high performance liquid chromatography in conjunction with two programmable fluorescence detectors. Sensitivity and selectivity were enhanced by analyzing PAHs under their optimum fluorescence wavelengths. The accuracy of the analytical method was evaluated by determination of PAHs in 5 mg of standard reference material SRM 1649. The method was also successfilly employed to analyze major parent PAHs and some alkyl-PAHs from environmental tobacco smoke [ETS] with a sample size of 2 mg using class-selective fluorescence wavelengths. Some alkyl-PAHs were tentatively identified even in the absence of standard compounds. Coeluting pairs were identified and analyzed by careful selection of excitation and emission wavelengths for each compound. Identities of the signals were confirmed by comparing both the retention behavior and the peak-height ratios at two or more different excitation and emission wavelength combinations.

Book excerpt: The combination of HPLC with phase-modulation spectrofluorometry provides fluorescence spectral and lifetime selectivity for the qualitative and quantitative analysis of Chromatographic peak components. The phase-modulation spectrofluorometer can be used for on-line HPLC detection and for batch analysis of Chromatographic fractions, as discussed in this paper for the analysis of mixtures of polycyclic aromatic hydrocarbons. In the case of on-line detection, fluorescence lifetime heterogeneity of Chromatographic peaks and peak portions is used as an indicator of Chromatographic peak purity. Fluorescence lifetimes and fractional intensity contributions can be determined from the observed phase and modulation lifetimes at an appropriate number of excitation modulation frequencies. Batch analysis of Chromatographic fractions can be performed by phase-resolved fluorescence spectroscopy, and involves the use of multidimensional data formats in which phase-resolved fluorescence intensity is plotted as a function of Chromatographic and fluorimetric experimental parameters. Advantages of the combined HPLC/phase-modulation spectrofluorometer system are discussed.
