Book excerpt: Bioremediation in the vadose zone is unpredictable because of poor understanding of factors influencing microbial growth in this environment. A lab-scale experimental system was developed to examine, noninvasively, interactions between microbial growth, water flow, and solute transport in unsaturated porous media. Measurements of microbial colonization, and its impact on hydrology, were facilitated by using the luxCDABE-containing reporter bacterium Pseudomonas fluorescens HK44 and digital CCD imaging. Experiments were conducted in glass-walled two-dimensional flow cells (45 x 50 x 1 cm) packed with silica sand. Several bioengineering problems associated with chamber design and function required solution before microbial experiments were successful. These included: choice of materials for chamber components; development of sterilization, packing, and inoculation protocols; and development of procedures for data collection and chamber maintenance during experiments lasting several days. Bacterial growth was mapped daily by quantifying development of salicylate-induced bioluminescence. A model relating the rate of increase in light emission after induction successfully predicted microbial densities over four orders of magnitude (R2=0.95) provided that sufficient oxygen for the bioluminescence reaction was available. Total model-predicted growth during a one-week experiment agreed with potential growth calculated from the mass-balance of the system and previously established kinetic parameters (predicted, 1.2x1012 cells; calculated, 1.7x1012 cells). Although the rate of expansion of the colonized zone (and predicted populations in newly colonized regions) remained relatively constant, the proportion of the daily potential growth remaining within the chamber declined over time. Monitoring of bioluminescence revealed the development of an (hypothesized) anaerobic zone associated with microbial growth in the unsaturated porous media. Water content and flow streams were measured using light transmission. Accumulation of microbial growth modified the hydrologic properties of the sand causing up to 50% decrease in saturation within the colonized zone, diversion of flow around the colonized zone, and lowering (5 cm) of the capillary fringe height. Apparent solute velocity through the colonized region was reduced from 0.39 cm min−1 (R2=0.99) to 0.25 cm min−1 (R2=0.99). These experiments provide proof-of-concept for combining light transmission and bioluminescence technologies to study interactions between microbial growth and hydrology in unsaturated porous media.

The effects can range from poor clarifier biomass settling and elevated effluent BOD and ammonia levels to total plant kills. These problems could be minimized or eliminated if an effective method existed for continuously monitoring biological wastewater treatment plant influent for toxicity to the treatment plant microorganisms. Current influent screening methods have not been proven to be adequate for adaptation to continuous screening in the field. The primary reasons include the batch-wise nature of the assays and an inadequate correlation between the assays and plant performance. The goal of the research team was to create new bioluminescent biosensors from different types of bacteria found in biological wastewater treatment plants for the development of a multi-channel continuous monitoring system. A system built from multiple biosensors would make it possible to differentiate between potential influent toxicity effects to different classes of bacteria (such as nitrifying and heterotrophic bacteria). The research team found it unexpectedly challenging to apply common microbiological transformation methods for laboratory strains to the wastewater treatment plant strains. The research team generated six new bioluminescent bioreporters from bacteria that are typical constituents of activated sludge. Of particular significance is a bioreporter developed using a Hyphomicrobium sp., which is a slow growing bacterium known to be present in significant numbers in some activated sludge plants. Of the six generated, initial bioluminescence and toxicity screening indicated that one strain (a Pseudomonad) was a particularly promising candidate due to its ease of cultivation and high light production. Further toxicity testing, however, determined that the response of the strain to 48 organic compounds and 8 metals commonly found in wastewater was similar to that of a previously created strain, Shk1 (also a Pseudomonad). Further work is therefore needed in the generation of appropriate biosensors and test conditions for populations not represented by the new heterotrophic biosensor.

Book excerpt: An experimental and numerical modeling investigation was conducted to study interactions between microbial dynamics and transport processes in variably saturated porous media. These interactions are important in a variety of applied problems such as water and wastewater treatment, bioremediation, and oil-field recovery operations. These processes and interactions also have great ecological significance, with global scale implications for carbon cycling in the environment and the related issue of climate change. Experiments were conducted under variably saturated flow conditions in columns and 2D light-transmission chambers packed with translucent quartz sand. A bioluminescent Pseudomonas fluorescens bacterium was utilized in the experiments and bioluminescence was used as a non-destructive measure of bacterial density and distribution. In the column experiments, pressure heads increased (became less negative) at all measured depths, but significant changes in apparent volumetric water contents were only observed in the upper 5 cm of the columns. Permeability was reduced by a factor of 40 within one week during growth on glucose. In the chamber experiments, aqueous-phase saturations decreased by 7-9% in the region of primary colonization and the capillary fringe dropped by 5 cm during the 6-day experiment. The colonized region expanded laterally by 15 cm and upward against the flow by about 7-8 cm. The desaturation phenomenon resulted in increased lateral spreading of solutes around the colonized region. A numerical model was developed and used to help interpret the experimental data. Water flow was modeled using the single-phase Richards equation. Solute and bacterial transport, cell growth, substrate consumption, and gas diffusion were modeled using advection-dispersion-reaction equations. Observed changes in saturations and pressure heads were reproduced approximately using fluid-media scaling to represent an apparent surface-tension lowering effect, which was assumed to be due to sorption of cells and/or biosurfactants at gas-liquid interfaces. Microbial dynamics, and substrate and oxygen consumption were represented using first-order reversible kinetics for cell attachment/detachment, and dual Monod-type kinetics for cell growth and substrate and oxygen consumption. Reasonably good matches were obtained between the observed and simulated results.
