Book Adhesion G Protein coupled Receptors

Download or read book Adhesion G Protein coupled Receptors written by Tobias Langenhan and published by Springer. This book was released on 2016-11-09 with total page 409 pages. Available in PDF, EPUB and Kindle. Book excerpt: Latest research on Adhesion GPCRs has unearthed surprising revelations about the events that govern the signal transduction of these receptor molecules and the cellular and organ requirements for these signals. Unexpected and unprecedented findings suggest that Adhesion GPCRs constitute a group of receptors that sense mechanical stimuli and transcode them into metabotropic signals through the action of a novel activation paradigm. Interdisciplinary efforts transcending many areas of biomedical research including pharmacology, physiology, genetics, cell biology, structural biology, biochemistry and bioinformatics were necessary to unveil these fundamental properties. The scientific leaders in the field that carried this research effort have teamed up here to provide a comprehensive overview of our current understanding, how Adhesion GPCRs signal and how these receptors shape organ structure and function.

Book excerpt: Upon completion of the human genome project over 800 G protein-coupled receptor 1 (GPCR) genes, subdivided into five categories, were identified. These receptors sense a diverse array of stimuli, including peptides, ions, lipid analogues, light and odour, in a discriminating fashion. Subsequently, they transduce a signal from the ligand–receptor complex into numerous cellular responses. The importance of GPCRs is further reflected in the fact that they constitute the most common target for therapeutic drugs across a 2 wide range of human disorders. Phylogenetic analysis of GPCRs produced the GRAFS classification system, which subdivides GPCRs into five discrete families: glutamate, rhodopsin, adhesion, frizzled/taste2 and secretin receptors. The adhesion-GPCR family 2 can be further subdivided into eight groups. The field of adhesion-GPCR biology has indeed become large enough to require a volume dedicated solely to this field. The contributors to this book have made a courageous effort to address the key concepts of adhesion-GPCR biology, including the evolution and biochemistry of adhesion-GPCRs; there are extensive discussions on the functional nature of these receptors during development, the immune response and tumourgenesis. Finally, there are chapters dedicated to adhesion-GPCR signalling, an area of intense investigation.

Book The Extracellular Regions of Adhesion G Protein Coupled Receptors Regulate Ligand Binding and Intracellular Signaling

Download or read book The Extracellular Regions of Adhesion G Protein Coupled Receptors Regulate Ligand Binding and Intracellular Signaling written by Gabriel Simon Salzman and published by . This book was released on 2017 with total page 219 pages. Available in PDF, EPUB and Kindle. Book excerpt: G protein-coupled receptors (GPCRs) have emerged as incredibly successful drug targets. Members of the adhesion GPCRs (aGPCR) family are characterized by diverse extracellular regions (ECRs), which play roles in cell adhesion, and mediate a subset of aGPCR functions in vivo. Though these receptors are implicated in myriad disease processes, there are no aGPCR-targeted therapeutics to date, due in large part to both the absence of well-behaving ligands as well as the technical challenges associated with mechanistic studies of aGPCR ECRs. We present a structural and functional study of the aGPCR GPR56/ADGRG1, a receptor critical for neurodevelopment and leukemia progression. To overcome many of the challenges mentioned above, we generated over thirty synthetic protein ligands, termed monobodies, that bind diverse epitopes across the ECR. Using a monobody crystallization chaperone, we solved the structure of the full ECR of GPR56, a first for any aGPCR, revealing the domain boundaries in the ECR as well as the identity and unique fold of the previously undefined N-terminal domain. We showed this domain regulates signaling and natural ligand binding in vitro, is deleted via alternative splicing, and mediates myelination in vivo. Additionally, we developed monobodies with stimulatory and inhibitory functions, demonstrating that ECR-targeted ligands can directly regulate aGPCR signaling, and are therefore valuable experimental reagents as well as lead-molecules for therapeutic development. Our results suggest an intricate, ECR-mediated molecular mechanism underlying aGPCR regulation. With the ultimate goal of combating aGPCR-mediated diseases, our findings will pave the way for targeted therapeutic development.

Book The G Protein Coupled Receptors Handbook

Download or read book The G Protein Coupled Receptors Handbook written by Lakshmi A. Devi and published by Springer Science & Business Media. This book was released on 2008-03-01 with total page 414 pages. Available in PDF, EPUB and Kindle. Book excerpt: A comprehensive survey of the many recent advances in the field of G protein-coupled receptors (GPCR). The authors describe the current knowledge of GPCR receptor structure and function, the different mechanisms involved in the regulation of GPCR function, and the role of pharmacological chaperones in GPCR folding and maturation. They also present new findings about how GPCR dimerization/oligomerization modifies the properties of individual receptors and show how recent developments are leading to significant advances in drug discovery, such as the detection of ligands for orphan GPCRs. Also discussed are the most recent developments that could lead to new drug discoveries: the role of GPCRs in mediating pain, the development of receptor-type selective drugs based on the structural plasticity of receptor activation, and the identification of natural ligands of orphan GPCRs (deorphanization) as possible drug targets.

Book Transmembrane Signaling Protocols

Download or read book Transmembrane Signaling Protocols written by Hydar Ali and published by Springer Science & Business Media. This book was released on 2008-02-05 with total page 354 pages. Available in PDF, EPUB and Kindle. Book excerpt: The previous edition of Transmembrane Signaling Protocols was published in 1998. Since then the human genome has been completely sequenced and new methods have been developed for the use of microarrays and proteomics to analyze global changes in gene expression and protein profiles. These advances have increased our ability to understand transmembrane signaling processes in much greater detail. They have also simultaneously enhanced our ability to determine the role of a large number of newly identified molecules in signaling events. In addition, novel video microscopy methods have been developed to image transmembrane signaling events in live cells in real time. In view of these major advances, it is time to update the previous edition. Because of the success of that volume, we have chosen to keep the essential character of the book intact. Introductory chapters from experts have been included to provide overall perspective and an overview of recent advances in signal transduction pathways. The individual chapters now include comp- hensive detailed methods, studies in genetically tractable systems, fluorescence microscopy in live single cells, ex vivo analysis of primary cells from tra- genic mice, as well as genomic and proteomic approaches to the analysis of transmembrane signaling events. We would like to express our deep gratitude to the coauthors of this publi- tion. We hope that Transmembrane Signaling Protocols, Second Edition will serve as a valuable resource for future progress in the study of signal transd- tion pathways.
