In 1978, the "International Conference on Physiological and Regulatory Funetions of Adenosine and Adenine Nueleotides," held in Banff, Canada, served as an effective catalyst in enabling scientists with differ ent points of view to exchange information and ideas on this subject. Since 1978, the amount of research on different as peets of adenosine metabolism, mechanism of action, transport, and function in various tissues (in both nor maI and abnormal states) has increased exponentially, and there was an urgent need for another meeting for updating information on this burgeoning subject. The focus of the meeting was extremely sharp, but the scope of the subject material was very broad. Scientists categorized as chemists, biochemists, physiologists, pharmacologists, and immunologists, working with cells and eelI fragments, with tissues such as heart, brain, musele, fat, kidney, and with human beings having immune deficiency diseases participated in the sym posium. They had an opportunity to air their differences, reinforce their areas of agreement, and, most important, they educated each other with respeet to research on adenosine; it is hoped that they also enriched their own research programs from the information they gained from their fellow participants.

Another thirty years were to pass before the possible physiological role of adenosine in coronary vasodilation was studied by Berne and others. Since then, there has been a tremendous increase in research into the actions of adenosine. Workers from many disciplines have employed a wide variety of techniques, since adenosine is a product of and a substrate for a number of metabolic pathways, is transported into cells, and acts at discrete receptor sites to modulate the activity of adenylate cyclase and to produce important actions on many cells and tissues including platelets, adipo cytes, heart, blood vessels, and other smooth muscles. International symposia on the actions of adenosine were held in 1978, 1981, and 1982, and the proceedings of these symposia have been published (Baer and Drummond, 1979; Daly et at., 1983; Berne et at., 1983). Since it is not the primary purpose of the present volume to review our current understanding of the nu merous actions of adenosine, these volumes should be consulted for such details. Rather, the present volume has been planned to provide both graduate students and investigators in pharmacology and related disciplines with a summary of some of the methods now available for the study of the actions of adenosine and, in particular, to highlight their possible uses and limitations.

Non-invasive NMR technologies have been developed to monitor in vivo the energy level of tissues based on determination of the concentrations of adenine nucleotides, phosphate and phosphate esters. Thus it became clear that the capacity of oxidative phosphorylation adapts itself to the ATP demand which changes continuously with the physiological state in various tissues. This is achieved by regulation of the enzyme activity as well as by regulation of its biogenesis. The reversible phosphorylation of proteins is re­ cognised as a major regulatory mechanism in eukaryotic as well as in prokaryotic cells for cellular signal transduction and control of gene expression, cell growth, differentation and oncogenesis. The same applies to the role played by cAMP. A further topic of growing interest concerns the discovery of the ATP binding cassette (ABC) superfamily of transport proteins which includes systems of primary importance in medicine such as the multi-drug resistance P glycoprotein, the cystic fibrosis transmembrane conductance regulator (CFTR) and the 70 kd peroxisomal membrane protein. Finally, much attention is being devoted in many laboratories to the molecular structure and role of ATP­ modulated channels.
