Book Methods Used in Adenosine Research

Download or read book Methods Used in Adenosine Research written by David Paton and published by Springer Science & Business Media. This book was released on 2013-06-29 with total page 385 pages. Available in PDF, EPUB and Kindle. Book excerpt: In their classic paper in 1929, Drury and Szent-Gyorgyi described a number of the important cardiovascular actions of adenosine. Another thirty years were to pass before the possible physiological role of adenosine in coronary vasodilation was studied by Berne and others. Since then, there has been a tremendous increase in research into the actions of adenosine. Workers from many disciplines have employed a wide variety of techniques, since adenosine is a product of and a substrate for a number of metabolic pathways, is transported into cells, and acts at discrete receptor sites to modulate the activity of adenylate cyclase and to produce important actions on many cells and tissues including platelets, adipo cytes, heart, blood vessels, and other smooth muscles. International symposia on the actions of adenosine were held in 1978, 1981, and 1982, and the proceedings of these symposia have been published (Baer and Drummond, 1979; Daly et at., 1983; Berne et at., 1983). Since it is not the primary purpose of the present volume to review our current understanding of the nu merous actions of adenosine, these volumes should be consulted for such details. Rather, the present volume has been planned to provide both graduate students and investigators in pharmacology and related disciplines with a summary of some of the methods now available for the study of the actions of adenosine and, in particular, to highlight their possible uses and limitations.

Book Adenosine and Adenosine Receptors

Download or read book Adenosine and Adenosine Receptors written by Michael Williams and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 522 pages. Available in PDF, EPUB and Kindle. Book excerpt: Historically, the major emphasis on the study of purinergic systems has been predominantly in the areas of physiology and gross pharmacology. The last decade has seen an exponential in crease in the number of publications related to the role of both adenosine and A TP in mammalian tissue function, a level of interest that has evolved from a more molecular focus on the identity of adenosine and A TP receptor subtypes and the search for selective ligands and development of radioligand binding assays by Fred Bruns and colleagues (especially that for A receptors) that played z a highly significant role in advancing research in the area. In the 60 years since adenosine was first shown to be a potent hypotensive agent, a considerable investment has been made by several pharmaceutical companies-including Abbott, Byk Gulden, Takeda, Warner-LambertlParke Davis, Boehringer Mann heim, Boehringer Ingelheim, Nelson/Whitby Research and CffiA Geigy-as well as John Daly's laboratory at the National Institutes of Health, to design new adenosine receptor ligands, and both agon ists and antagonists with the aim of developing new therapeutic entiities. Numerous research tools have derived from these efforts including: 2-chloroadenosine, R-PIA (~-phenylisopropyladeno sine; NECA (5' N-ethylcarboxamidoadenosine); CV1808; CI936; PD 125,944; ~-benzyladenosine; PACPX; CPX; CPT; XAC; CGS 15943 and CGS 21680. Yet in the realm of therapeutics it was only in 1989 that adenosine itself was approved for human use in the treatment of supraventricular arrythmias.

Book Nucleosides Nucleotides and their Biological Applications

Download or read book Nucleosides Nucleotides and their Biological Applications written by Janet L Rideout and published by Academic Press. This book was released on 2012-12-02 with total page 340 pages. Available in PDF, EPUB and Kindle. Book excerpt: Nucleosides, Nucleotides, and Their Biological Applications contains the proceedings of the Fifth International Round Table on Nucleosides, Nucleotides, and Their Biological Applications held at Research Triangle Park, North Carolina on October 20-22, 1982. Contributors focus on the biological applications of nucleosides and nucleotides, along with advances in synthetic chemistry. Topic range from fast-moving biochemical subjects such as the 2',5'-oligoadenylates and their relationship to interferons; adenosine and adenine nucleotide receptors; and nucleosides with selective antiviral action. This volume is organized into 11 chapters and begins with an overview of nucleosides that are used as antiviral agents and their mechanism of action, including idoxuridine, vidarabine, and trifluridine. The discussion then shifts to the chemical and biological properties of nucleosides of purines and ring analogs; 2'-fluoroarabinosyl pyrimidines and purine-like C-nucleosides; and the potential of 2',5'-oligoadenylates as chemotherapeutic agents. The reader is also introduced to receptors for adenosine and adenine nucleotides; the function of pyrrolo[2,3-d]pyrimidine nucleosides in polynucleotides; and unusual nucleoside synthons and oligonucleotide synthesis. A chapter on the synthesis of versatile C-nucleoside precursors and certain C-nucleosides concludes the book. This book will be of value to chemists, biologists, and those with an interest in nucleosides and nucleotides.
