A noninvasive and pain free method of generating nerve block with magnetic stimulation would have many important clinical benefits. One method of electrical stimulation developed for creating nerve block is that of "collision block." This dissertation addresses how computer modeling with validation might be used to implement unidirectional action potentials (a necessary first step in implementing collision block) via the application of large time-varying magnetic fields. Specifically, the dissertation addresses condition numbers associated with the low frequency impedance modeling method. This was necessary to ensure computational accuracy in the predicted electric field gradients that result from the application of time-varying magnetic fields. Equations are provided that describe how many significant figures might be lost in computing an impedance method solution as a result of problem size, tissue properties, and implementation strategies. A constrained optimization technique derived from the impedance method is then described and used to determine an optimal coil design for delivering depolarizing and hyperpolarizing electric field gradients at closely spaced locations for the nerve. Although the coil design presented here was specifically meant for generating unidirectional action potentials, the constrained optimization technique permits specifying other stimulus constraints at one or multiple locations in tissue and can thus be used for many other neural stimulation paradigms. The coil designed for generating unidirectional action potentials using the constrained optimization technique was experimentally evaluated and computational and experimental results were in good agreement. Additional computer models were then used in conjunction with a coil of similar design to predict necessary conditions for generating unidirectional action potentials in mammalian peripheral nerve. A physical test arrangement based on these results was constructed and used to test whether unidirectional action potentials could be generated by magnetically induced activation and block of action potentials in mammalian peripheral nerve. Quantification of compound action potentials indicated that unidirectional action potentials occurred in some axons under these conditions.
