Book excerpt: Malignant pleural mesothelioma (MPM) is a rare thoracic cancer that derives from the mesothelial cells of the pleura and is causally associated with exposure to asbestos. Because of its aggressiveness and resistance to therapies, MPM prognosis is extremely poor and the 5-year survival rate is approximately 10%. Despite its poor effectiveness, chemotherapy with cisplatin and pemetrexed has long remained the standard-of-care for upfront treatment with no second line therapy available. Recently, the approval of ipilimumab (anti-CTLA-4) plus nivolumab (anti-PD-1) for the first-line treatment of unresectable MPM has marked a significant milestone, in particular for the non-epithelioid MPM subtype, which is more resistant to chemotherapy and associated with a poorer outcome. However, most MPM patients are still resistant to cancer therapies. Beyond histologic subtypes, recent studies have highlighted extensive genetic variation and gene expression deregulation both between and within MPM patients. This molecular heterogeneity highlights the need for a personalized treatment approach. Therefore, both multiple therapeutic options and predictive biomarkers that can guide clinical decision-making are urgently needed. Tumor cell immunogenicity and tumor microenvironment composition (TME) are the major determinants of the success of immunotherapy. Despite the low tumor mutational burden, recent studies indicate that chromoanagenesis and chromotripsis are molecular features of MPM. Since chromosomal instability can lead to the expression of neo-antigens, it could also influence the efficacy of immune checkpoint inhibitors (ICIs). The immunosuppressive TME is an additional important hurdle that limits the efficacy of immunotherapy. The combination of ICIs with strategies to boost anti-tumor immunity by restraining immunosuppressive myeloid cells or by reprograming “cold tumor” into “hot tumor” is an active area of research. This Research Topic aims to discuss recent advances in understanding and targeting MPM immune microenvironment for therapeutic purposes along with potential predictive biomarkers to select patients who most likely will respond to immunotherapies. We are interested in basic and clinical studies that explore the crosstalk between tumor and immune cells in order to highlight new molecular targets, approaches, and combination treatments to enhance anti-tumor immunity in MPM and assess potential predictive biomarkers of response to immunotherapy.

Needless to say, my views reflect the biases that I have acquired. One way to articulate the rationale (and limitations) of my biases is through the preface of a truly great text of a previous era, Cooley and Lohnes (1971, p. v). They draw a distinction between mathematical statisticians whose intel lect gave birth to the field of multivariate analysis, such as Hotelling, Bartlett, and Wilks, and those who chose to "concentrate much of their attention on methods of analyzing data in the sciences and of interpreting the results of statistical analysis . . . . (and) . . . who are more interested in the sciences than in mathematics, among other characteristics. " I find the distinction between individuals who are temperamentally "mathe maticians" (whom philosophy students might call "Platonists") and "scientists" ("Aristotelians") useful as long as it is not pushed to the point where one assumes "mathematicians" completely disdain data and "scientists" are never interested in contributing to the mathematical foundations of their discipline. I certainly feel more comfortable attempting to contribute in the "scientist" rather than the "mathematician" role. As a consequence, this book is primarily written for individuals concerned with data analysis. However, as noted in Chapter 1, true expertise demands familiarity with both traditions.

In contrast to the social-psychological approach to studying voting behaviour, the spatial theory of voting is premised on the idea of self-interested choice. Voters cast votes on the basis of their evaluation of the candidates or policy alternatives competing for their vote. Candidates fashion their appeals to the voters in an effort to win votes. The spatial theory provides explicit definitions for these behavioural assumptions to determines the form that self-interested behaviour will take. The consequences of this behaviour for the type of candidate or policy that voters will select is the major focus of the theory. There is a twofold purpose to this work. The first is to provide an elementary but rigourous introduction to an important body of political science research. The second is to design and test a spatial theory of elections that provides insights into the nature of election contests. The book will appeal to a wide audience, since the mathematics is kept to an accessible level.

He was the principal author of The Rehearsal (1671), an enormously successful burlesque play that ridiculed John Dryden and the rhymed heroic drama. Historians remember Buckingham as an opponent who helped topple Clarendon from power in 1667, as a member of the 'Cabal' government in the early 1670s, and as an ally of the Earl of Shaftesbury in the political crisis of 1678-1683. The duke was prominent among the 'court wits' (Rochester, Etherege, Sedley, Dorset, Wycherley, and their circle); he was closely associated with such writers as Butler and Cowley; he was a conspicuous champion of religious toleration and a friend of William Penn. No edition of Buckingham has been published since 1775, partly because his work presents horrendous attribution problems. He was (probably) adapter or co-author of six plays (two of them vastly successful for more than a century) including one in French that appears here in English for the first time. He is also associated with nine topical pieces (variously political, religious, and satiric) and some twenty poems of wildly varying type. The 'Buckingham' commonplace book has previously been published only in fragmentary form. Almost all of these works present major difficulties in both attribution and annotation, here seriously addressed for the first time. This edition is a companion venture to Harold Love's important edition of Rochester (OUP, 1999).
