Contamination of soil and water with toxic levels of zinc has been a growing problem since the Industrial Revolution. Historic mining in the Coeur d' Alene, Idaho area resulted in zinc levels of over 6000 ppm in local river bank and lake bottom soils. Soil bacteria are able to resist the toxicity of zinc and other metals in several ways that reduce metal mobility and bioavailability. The goal of this study was to develop methods for selection, characterization and genetic analysis of soil bacteria that accumulate zinc. Soil samples were collected from the zinc-laden banks of the Coeur d'Alene River and a non-contaminated site on the Eastern Washington University campus. The physical characteristics, zinc content, and native bacterial populations in the soils were analyzed. Soil microbial communities were selected and stimulated for high zinc tolerance by culture in 50-mL Zn/Cu electrolytic cell bioreactors. Highly zinc tolerant clones were selected from the resulting consortia on ZnSO4 gradient agar petri dishes. The zinc tolerance phenotypes of both native and in-vitro selected bacteria were studied on a novel microtiter plate zinc tolerance assay. Quantitative measures of zinc tolerance included the zinc minimum inhibitory concentration (MIC) and concentration inhibiting growth to 50% (IC50). The zinc accumulation capacity in eight zinc tolerant isolates was measured by growing pure cultures in broth with 300 ppm zinc, harvesting and washing the bacteria, then determining the zinc content by flame atomic absorption. The genes responsible for zinc tolerance and accumulation were sought through plasmid extractions and polymerase chain reaction (PCR) screening with primers for the gene encoding a known zinc-binding protein (PZPl). The zinc-contaminated Coeur d' Alene soils contained only a fraction of the culturable microbes observed in the campus soil.
