Water is critically important to the structure and function of biological membranes. A detailed understanding of the behavior of water in membranes is thus important to under- standing the basic mechanisms of life. Nitrous oxide may be employed as a probe of water in membranes. In this work, we study the physics of water and nitrous oxide (N2 O ) in model membranes using classical molecular dynamics and assess the ability of simple water models to capture phase transitions. Phospholipid bilayers are a major component of cell membranes. We perform atomistic-scale simulations of phospholipid bilayers at three hydrations. These simulations offer a detailed picture of both water structure and dynamics, allowing us to study the properties of water as a function of hydration with high spatial resolution. We find an interesting crossover phenomenon in the diffusion of water at the phosphorus density maximum regardless of the overall hydration level. While most properties do not change beyond the commonly established excess water point, the water diffusion coefficient still increases upon further hydration. We propose a scheme that classifies lipid hydration water into buried water, interface water, and bulk-like water.We also use atomistic-scale simulations to calculate the free-energy profile of N2O in a lipid bilayer. The free-energy profile gives information about binding sites, partition coefficients of N2O in lipid and water, and the activation barrier for N2O penetration both into and across the lipid. This information is valuable for understanding the anesthetic action of N2O and its role as a probe molecule. The adaptive force matching method provides a mechanism to assess the effectiveness, quality, and transferability of a computational model. We apply this method to create high-quality models for liquid water and ice, assess the ability of one model to capture both phases, and calculate the melting points of the models by computing the free energy difference between phases. The free energy difference is zero at the melting point, a first-order phase transition. Such a computation is numerically challenging. We construct a novel thermodynamic pathway that avoids a first-order phase transition.
