This volume describes augmentation approaches, where improvements in brain functions are achieved by modulation of brain circuits with electrical or optical stimulation, or pharmacological agents. Activation of brain circuits with electrical currents is a conventional approach that includes such methods as (i) intracortical microstimulation (ICMS), (ii) transcranial direct current stimulation (tDCS), and (iii) transcranial magnetic stimulation (TMS). tDCS and TMS are often regarded as noninvasive methods. Yet, they may induce long-lasting plastic changes in the brain. This is why some authors consider the term “noninvasive” misleading when used to describe these and other techniques, such as stimulation with transcranial lasers. The volume further discusses the potential of neurostimulation as a research tool in the studies of perception, cognition and behavior. Additionally, a notion is expressed that brain augmentation with stimulation cannot be described as a net zero sum proposition, where brain resources are reallocated in such a way that gains in one function are balanced by costs elsewhere. In recent years, optogenetic methods have received an increased attention, and several articles in Volume II cover different aspects of this technique. While new optogenetic methods are being developed, the classical electrical stimulation has already been utilized in many clinically relevant applications, like the vestibular implant and tactile neuroprosthesis that utilizes ICMS. As a peculiar usage of neurostimulation and pharmacological methods, Volume II includes several articles on augmented memory. Memory prostheses are a popular recent development in the stimulation-based BMIs. For example, in a hippocampal memory prosthesis, memory content is extracted from hippocampal activity using a multiple-input, multiple-output non-linear dynamical model. As to the pharmacological approaches to augmenting memory and cognition, the pros and cons of using nootropic drugs are discussed.

Humans possess the ability to recognise thousands of people very quickly and accurately without effort. The serious social difficulties that follow abnormalities of the face recognition system (i.e., prosopagnosia) strongly underline the importance of typical face skills in our everyday life. Over the last fifty years, research on prosopagnosia, along with research in the healthy population, has provided insights into the cognitive and neural features behind typical face recognition. This has also been achieved thanks to non-invasive neuroimaging techniques such as functional Magnetic Resonance Imaging (fMRI), Electroencephalography (EEG), Magnetoencephalography (MEG), Diffusion Tensor Imaging (DTI) and Transcranial Magnetic Stimulation (TMS). However, there is still much debate about the cognitive and neural mechanisms of face perception. In the current “Research Topic” we plan to gather experimental works, opinions, commentaries, mini-reviews and reviews that focus on new or novel theories and methods in face perception research. Where is the field at the moment? Do we need to re-think the experimental procedures we have adopted so far? Again, what kind of techniques (or combination of them) and analysis methods will be important in the future? From the experimental point of view we encourage both behavioural and neuroimaging contributions (e.g., fMRI, EEG, MEG, DTI and TMS). Despite the main emphasis on face perception, memory and identification, we will also consider original works that focus on other aspects of face processing, such as expression recognition, attractiveness judgments and face imagery. In addition, animal investigations and experimental manipulations that alter face recognition abilities in typical human subjects (e.g., hypnosis) are also welcome. Overall, we are proposing a Research Topic that looks at face processing using different perspectives and welcome contributions from different domains such as psychology, neurology, neuroscience, cognitive science and philosophy. The current “Research Topic” evolved over the desire to acknowledge the relatively recent loss of three giants in the field: Drs. Shlomo Bentin, Truett Allison and Andy Calder. We dedicate this “Research Topic” to them and their pioneering studies.

As a consequence, the overall process leading to the production of a successful drug is very costly. The improvement of the net outcome in drug discovery and development would require, amongst other important factors, a good understanding of the molecular events that characterize the disease or pathology in order to better identify likely targets of interest, to optimize the interaction of an active agent (small molecule or macromolecule of natural or synthetic origin) with those targets, and to facilitate the study of the pharmacokinetics, pharmacodynamics and toxicity of an active agent in suitable models and in human subjects. The objective of this Research Topic is to highlight new developments and applications of imaging techniques with the objective of performing pharmacological studies in vivo, in animal models and in humans. In the domain of drug discovery, the pharmacological and biomedical questions constitute the center of attention. In this sense, it is fundamental to keep in mind the strengths and limitations of each analytical or imaging technique. At the end, the judicious application of the technique with the aim of supporting the search for answers to manifold questions arising during a long and painstaking path provides a continuous role for imaging within the complex area of drug discovery and development.

Book excerpt: In the last few years, advances in human structural and functional neuroimaging (fMRI, PET, EEG/MEG) have resulted in an explosion of studies investigating the anatomical and functional connectivity between different regions of the brain. More and more studies have employed resting and task-related connectivity analyses to assess functional interactions, and diffusion-weighted tractography to study white matter organization. Many of these studies have addressed normal human function, but recently, a number of investigators have turned their attention to examining brain disorders. The study of brain disorders is a complex endeavor; not only does it require understanding the normal brain, and the regions involved in a particular function, but also it needs a deeper understanding of brain networks and their dynamics. This Research Topic will provide the scientific community with an overview of how to apply connectivity methods to study brain disease, and with perspectives on what are the strength and limitations of each modality. For this Research Topic, we solicit both reviews and original research articles on the use of brain connectivity analysis, with non-human or human models, to explore neurological, psychiatric, developmental and neurodegenerative disorders from a system perspective. Connectivity studies that have focused on one or more of the following will be of particular interest: (1) detection of abnormal functional/structural connectivity; (2) neural plasticity, assessed by changes in connectivity, in patients with brain disorders; (3) assessment of therapy using connectivity measures; (4) relation of connectivity changes to behavioral changes.

This challenge will only be accomplish by a multidisciplinary approach involving genetics, molecular biology, optics, ethology, neurobiology and mathematics and using tractable model systems. The zebrafish larva is a transparent genetically tractable small vertebrate, ideal for the combination state-of-the- art imaging techniques (e.g. two-photon scanning microscopy, single-plane illumination microscopy, spatial light modulator microscopy and lightfield microscopy), bioluminiscence and optogenetics to monitor and manipulate neuronal activity from single specific neurons up to the entire brain, in an intact behaving organism. Furthermore, the zebrafish model offers large and increasing collection of mutant and transgenic lines modelling human brain diseases. With these advantages in hand, the zebrafish larva became in the recent years, a novel animal model to study neuronal circuits and behaviour, taking us closer than ever before to understand how the brain controls behaviour.
